Identification of bluetongue virus serotypes in farms of the Cuba western region

Bluetongue (BT) is an insect-transmitted viral disease of ruminant species. It is listed as a “notifiable disease” by the Mundial Organization for Animal Health. In Cuba, only we have knowledge about circulation of the virus by serological evidence and viral isolation but it has not been possible to carry out its molecular typing. The present work was aimed to know the serotypes of bluetongue virus (BTV) circulating in western region of Cuba. Five bovine units located in that region of the country were selected from Havana and Mayabeque provinces. Blood from young bovine and ovine were collected. Samples were placed in EDTA bottles and stored at -4°C until processing at Virology laboratory in France. Extraction of RNA was carried out from the collected blood for identification by RT-PCR of BTV and identification of the serotype by real-time RT-PCR. Finally, were obtained virus isolates for an in-depth characterization of the circulating virus strains by molecular biology techniques such as next-generation sequencing, using high-tech equipment. These results constitute an ideal approach in order to development strategies for the surveillance and control of this disease in the country.
